Drug intelligence / Profile preview
The TNFSF8-CpG vaccine (also known as the TNFSF8-targeting CpG peptide vaccine) is an experimental therapeutic peptide vaccine designed to prevent immune checkpoint blockade (ICB)-induced pneumonitis (CIP) while synergistically enhancing the anti-tumor efficacy of cancer immunotherapy. Developed by researchers at Nanfang Hospital, Southern Medical University, the vaccine targets the extracellular domain of tumor necrosis factor ligand superfamily member 8 (TNFSF8, also known as CD30 ligand or CD153). TNFSF8+ CD4+ T cells have been identified as key lung-specific effectors driving CIP through a CD30-TNFSF8-ADGRE5-SFTPD lung-homing axis, leading to neutrophil recruitment and activation. The vaccine is formulated with a CpG oligodeoxynucleotide (specifically CpG ODN 1585) adjuvant, which stimulates Toll-like receptor 9 (TLR9) to promote Th1 polarization. In preclinical models, the vaccine successfully induces TNFSF8-specific antibodies, selectively depletes circulating TNFSF8+ CD4+ T cells, prevents lung injury and fibrosis, and boosts the anti-tumor efficacy of immune checkpoint inhibitors.
