TNG961 is a first-in-class, potent, and selective molecular glue degrader that targets HBS1L for the treatment of cancers with FOCAD deletion. FOCAD deletion occurs in 20-40% of all MTAP-deleted cancers and is particularly common in non-small cell lung cancer (NSCLC), affecting approximately 7% of these patients. Cancers with FOCAD deletion are dependent on HBS1L for mRNA processing and protein synthesis due to destabilization of the SKI complex. By degrading HBS1L and disrupting the HBS1L/PELO complex, TNG961 induces tumor regression in preclinical models across multiple histologies. The drug is being developed as an oral therapy by Tango Therapeutics for genetically defined subsets of solid tumors with limited treatment options[1][3][4].
