Overview

TNP (N2-(m-(trifluoromethyl)benzyl), N6-(p-nitrobenzyl)purine) is a potent, cell-permeable small molecule inhibitor of inositol hexakisphosphate kinases (IP6Ks), with a primary focus on IP6K1. Originally developed as a research tool to investigate the signaling roles of inositol pyrophosphates such as IP7, TNP has more recently been studied for its therapeutic potential in metabolic and hematologic conditions. In the context of oncology, TNP is used to block the IP6K1-driven synthesis of inorganic polyphosphates (polyP). Research suggests that tumor-derived polyP, often released via extracellular vesicles, acts as a procoagulant surface that triggers the contact activation system. By inhibiting IP6K1, TNP reduces polyP production and may mitigate the risk of immune checkpoint inhibitor-associated thrombosis (IAT) and other forms of cancer-associated thrombosis.
