Overview

TNP-470 is a synthetic analog of fumagillin, an antibiotic originally isolated from the fungus Aspergillus fumigatus. It functions as a potent angiogenesis inhibitor with antineoplastic activity. The drug binds to and irreversibly inactivates methionine aminopeptidase 2 (MetAP2), leading to endothelial cell cycle arrest in late G1 phase and inhibition of tumor angiogenesis. This mechanism results in suppression of new blood vessel formation required for tumor growth and metastasis. TNP-470 has been investigated primarily for cancer indications, including solid tumors, Kaposi's sarcoma, pancreatic neoplasms, renal cell carcinoma, and others[1][2][5]. Preclinical studies also suggest effects on adipose tissue growth and metabolic parameters[7].
