Overview

TNP-Cat-B is a tumor-enzyme activatable theranostic nanoprobe developed for the treatment and imaging of glioblastoma (GBM). It comprises ferumoxytol nanoparticles conjugated to the cytotoxic tubulin inhibitor monomethyl auristatin E (MMAE) via a linker that is specifically cleaved by the enzyme cathepsin B, which is overexpressed in glioblastoma cells. This design allows for targeted drug release within the tumor microenvironment, minimizing systemic toxicity. Additionally, the ferumoxytol component acts as a contrast agent for magnetic resonance imaging (MRI), allowing for the non-invasive monitoring of drug delivery and therapeutic efficacy through T2* relaxation time measurements. In preclinical studies, TNP-Cat-B has been evaluated as a monotherapy and in combination with TNP-MMP-14, a vascular-disrupting nanoprobe intended to open the blood-brain barrier to maximize delivery.
