Overview

TNP-MMP-14 is a tumor-enzyme activatable theranostic nanoprobe designed for the treatment and imaging of glioblastoma. It is specifically engineered to be activated by Matrix metalloproteinase-14 (MMP-14), an enzyme overexpressed in glioblastoma tissues but not in normal brain tissue. Upon activation, the nanoprobe disrupts the blood-tumor barrier (BTB), facilitating enhanced drug delivery to the tumor site. As a theranostic agent, it enables magnetic resonance imaging (MRI) guidance, allowing for the monitoring of tumor accumulation and therapeutic response. In preclinical studies, it has been evaluated in combination with other enzyme-activatable probes, such as TNP-Cat-B, to improve therapeutic efficacy and overcome drug delivery challenges in brain tumors.
