TNRX-257 is a novel multifunctional biologic developed by Tentarix Biotherapeutics using its proprietary Tentacles™ platform, which utilizes fully human stabilized antibody VH domains. The molecule is designed to simultaneously block the LAG3 (Lymphocyte-activation gene 3) checkpoint and conditionally deliver IL-2 receptor gamma/beta (IL2Rγ/β) agonism specifically to LAG3-expressing cells. This targeted approach aims to activate and expand tumor-reactive T cells within the tumor microenvironment while minimizing the systemic toxicity and pleiotropic effects typically associated with IL-2 therapy. TNRX-257 acts as a partial agonist for the IL-2 receptor, a profile that helps preserve the stemness of CD8+ T cells and preferentially activates effector T cells over regulatory T cells (Tregs). Preclinical data in humanized mouse models of melanoma and colorectal cancer have demonstrated robust anti-tumor efficacy and favorable pharmacokinetic properties, supporting its continued development for oncology indications.
