**TNX-1600** is a novel triple reuptake inhibitor (TRI) that simultaneously inhibits the reuptake of three key neurotransmitters: **serotonin**, **norepinephrine**, and **dopamine**. It is based on a unique pyran molecular scaffold and demonstrates nanomolar potency at all three monoamine transporters (NET > SERT ≈ DAT), with little to no affinity for other central nervous system receptors. TNX-1600 has shown robust antidepressant activity and normalization of traumatic stress-induced extinction-retention learning in animal models, with greater efficacy relative to paroxetine. Its primary indication is for the treatment of **posttraumatic stress disorder (PTSD)**, and it may have potential in other central nervous system disorders. It is currently in preclinical development and has not been approved for any indication[1][3][5][7][11].
