TNX-1700 is a stabilized recombinant version of Trefoil Factor 2 (TFF2), developed as a biologic candidate for the treatment of gastric and colorectal cancers. TFF2 is a small secreted protein expressed in gastrointestinal mucosa that plays roles in protecting and repairing mucosal tissue. In cancer, particularly gastric cancer, TFF2 expression is often epigenetically silenced; restoring its function may help suppress tumor development through several mechanisms. TNX-1700 targets the tumor microenvironment by reducing immunosuppressive myeloid-derived suppressor cells (MDSCs), especially polymorphonuclear MDSCs (PMN-MDSCs), thereby enhancing anti-tumor immune responses—most notably when combined with anti-PD1 checkpoint inhibitor therapy. Preclinical studies have shown that TNX-1700 can activate CD8+ T cells and reduce immune evasion by tumors, potentially converting anti-PD1 non-responsive tumors into responsive ones[1][4][6][7][8].
