TNX-1800 is a live attenuated modified horsepox virus vaccine designed to express the Spike protein of the SARS-CoV-2 virus and to elicit a predominant T cell immune response. It is administered percutaneously using a two-pronged ("bifurcated") needle. The platform leverages the immunogenicity and safety profile of horsepox, which is closely related to vaccinia (the smallpox vaccine vector), but with substantially decreased virulence in animal models. TNX-1800 has shown promise in preclinical studies for protecting animals from SARS-CoV-2 challenge and inducing both neutralizing antibodies and functional T cell immunity. The "take" reaction at the vaccination site serves as a biomarker for protective immunity, similar to historical smallpox vaccination practices[1][3][7].
