TNX-1900 is a proprietary, investigational drug-device combination product consisting of a novel intranasal formulation of oxytocin with added magnesium, designed to enhance the potency and specificity of oxytocin at its receptor. The formulation is delivered via an actuator device directly into the nasal cavity, targeting the trigeminal system. Oxytocin acts as a peptide hormone and neurotransmitter in the brain; magnesium in the formulation further potentiates its action at oxytocin receptors and may reduce paradoxical high-dose inhibition. The mechanism includes binding to neuronal oxytocin receptors in the trigeminal ganglia, inhibiting release of calcitonin gene-related peptide (CGRP), which plays a key role in migraine pathogenesis and pain signaling. This multimodal approach aims to provide analgesic effects for chronic migraine prevention while limiting systemic exposure and off-target effects. TNX-1900 is being developed for several indications including chronic migraine prevention, obesity (including adolescent obesity), binge eating disorder, social anxiety disorder (SAD), autism spectrum disorder (ASD)–related bone health issues, craniofacial pain syndromes, insulin resistance, and has also been studied for other pain conditions[1][2][5][6][7].
