Overview

TNX-2900 is a proprietary, investigational formulation of intranasal oxytocin potentiated with magnesium (Mg2+), developed by Tonix Pharmaceuticals for the treatment of Prader-Willi syndrome (PWS) in children and adolescents. Oxytocin is a naturally occurring hormone that acts as an anorexigenic agent—reducing appetite and signaling fullness—by activating oxytocin receptors in the brain. The addition of magnesium enhances the potency and specificity of exogenous oxytocin for its receptor, potentially improving therapeutic efficacy compared to non-potentiated formulations. Preclinical studies suggest that this Mg2+-potentiation increases receptor binding and may address limitations seen with standard intranasal oxytocin in PWS clinical trials. TNX-2900 has received Orphan Drug designation (2022) and Rare Pediatric Disease Designation (2024) from the FDA for PWS, which is characterized by severe hyperphagia leading to life-threatening childhood obesity[1][2][3][4][5][6][7][8][9].
