Overview

TNX-833 is a humanized monoclonal antibody developed by Tonix Pharmaceuticals that targets the CD40 ligand (CD40L), also known as CD154. CD40L is a costimulatory molecule primarily expressed on the surface of activated T cells, and its interaction with the CD40 receptor on B cells and other antigen-presenting cells is essential for the activation of the adaptive immune response. TNX-833 acts as an antagonist, blocking this interaction to inhibit T-cell-mediated immune responses and the production of pathogenic antibodies. This mechanism is being explored for the prevention of organ transplant rejection and the treatment of autoimmune disorders. Notably, TNX-833 is engineered with a modified Fc region designed to minimize the risk of thromboembolic complications, a safety issue that previously halted the development of earlier anti-CD40L antibodies.
