Overview

TO-115 is a synthetic small molecule anti-nephritic agent, chemically identified as 4-chloro-3',6'-dimethyl-2,2'-iminodibenzoic acid. Developed by Taisho Pharmaceutical, it has been investigated for its potential in treating autoimmune kidney diseases, such as lupus nephritis. In preclinical studies using MRL/lpr mice (a model for systemic lupus erythematosus), TO-115 demonstrated the ability to restrain clinical symptoms of autoimmune kidney disease, including reducing proteinuria and blood urea nitrogen (BUN) levels, and improving survival time. Notably, its therapeutic effect appears to be independent of systemic immunosuppression; unlike conventional agents such as cyclophosphamide, TO-115 does not significantly suppress the production of autoantibodies or immune complexes. This suggests a mechanism of action focused on direct renal protection or local anti-inflammatory effects rather than broad immune modulation.
