Overview
TO-2070 is a novel, rapidly absorbed intranasal formulation of **dihydroergotamine mesylate (DHE)** developed as a drug-device combination for the **treatment of acute migraine**. It utilizes proprietary nasal powder drug delivery technology from Shin Nippon Biomedical Laboratories (SNBL) to enable faster onset of action compared to existing DHE treatments. DHE is an ergot alkaloid that acts as a **serotonin receptor agonist**, primarily targeting 5-HT1B/1D receptors to constrict cranial blood vessels and inhibit neurogenic inflammation in migraine pathophysiology. The program was led by **Transcept Pharmaceuticals**, with development later transitioning to **Satsuma Pharmaceuticals**.[1][7][10][19]
