TOAD-1 is a next-generation antibody-drug conjugate (ADC) developed by T.O.A.D. Oncology S.A. for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The drug consists of a monoclonal antibody specifically engineered to target Prostate-Specific Membrane Antigen (PSMA), a cell-surface protein that is significantly overexpressed in prostate cancer cells compared to normal tissues. Upon binding to PSMA, the ADC is internalized into the cancer cell through receptor-mediated endocytosis. Once inside the lysosomal compartment, the cytotoxic payload is released, typically leading to the disruption of microtubule dynamics or DNA damage, which ultimately triggers cell cycle arrest and apoptosis. TOAD-1 is designed to optimize the therapeutic index by utilizing a stable linker and a potent payload, aiming to reduce the systemic toxicities often associated with earlier PSMA-targeted ADCs while maintaining high anti-tumor efficacy in advanced disease settings.
