Tobemstomig is a bispecific monoclonal antibody designed to target and bind both programmed cell death protein 1 (PD-1) and lymphocyte activation gene 3 protein (LAG-3, CD223) on T cells. By inhibiting the PD‑1 and LAG‑3 immune checkpoint pathways, which are often exploited by tumors to evade immune surveillance, tobemstomig reinvigorates T-cell activation and proliferation. This dual blockade enhances cytotoxic T lymphocyte activity against tumor cells, aiming to restore antitumor immunity. Tobemstomig is being developed primarily as an immunotherapy for various advanced or metastatic solid tumors, including triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), renal cell carcinoma (RCC), melanoma, esophageal squamous cell carcinoma, urothelial cancer, and hepatocellular carcinoma. It has been evaluated both as monotherapy and in combination with chemotherapy or other immunotherapies in clinical trials[2][5][6][8].
