Toca-1 shRNA is a short hairpin RNA (shRNA) designed to knock down the expression of Transducer of Cdc42-dependent actin assembly-1 (Toca-1, also known as formin-binding protein 1-like or FNBP1L). Toca-1 is an adaptor protein that promotes the recruitment of actin nucleation-promoting factors to cellular membranes, facilitating cell motility and tissue invasion. Delivered via lentiviral vectors, Toca-1 shRNA has been utilized in preclinical research to investigate the role of Toca-1 in breast cancer metastasis. In vitro and in vivo studies demonstrate that Toca-1 knockdown significantly impairs EGF-induced cell migration, invasion, and extracellular matrix degradation, leading to a reduction in spontaneous lung metastasis without affecting primary tumor growth.
