Tocofersolan is a synthetic, water-soluble derivative of vitamin E (α-tocopherol) created by conjugating it with polyethylene glycol succinate. Unlike natural forms of vitamin E, which are fat-soluble, tocofersolan’s amphipathic structure allows it to form micelles and be absorbed in the absence of bile salts. This property makes it effective for treating vitamin E deficiency in patients with fat malabsorption disorders such as congenital or hereditary chronic cholestasis. It is approved in the European Union under the brand name Vedrop for pediatric patients with these conditions. Tocofersolan also acts as an antioxidant and has applications as an excipient and absorption enhancer in pharmaceuticals and cosmetics[1][2][3][5]. Its mechanism involves halting lipid peroxidation by acting as a free radical scavenger, thereby protecting cell membranes from oxidative damage[2]. Additionally, it can inhibit P-glycoprotein (ABCB1) activity and tyrosine kinase activity according to some biochemical studies[6].
