Tocopheryl phosphate, specifically the Tocopheryl Phosphate Mixture (TPM®), is a proprietary drug delivery platform developed by Phosphagenics (now Avecho Biotechnology). It consists of a mixture of mono-tocopheryl phosphate and di-tocopheryl phosphate, which are phosphorylated derivatives of Vitamin E. TPM acts as a lipid-based delivery system that enhances the solubility and absorption of poorly water-soluble drugs across biological membranes, including the skin and gastrointestinal tract. Beyond its role as a carrier, it exhibits intrinsic anti-inflammatory properties by modulating pro-inflammatory cytokines such as IL-1β, IL-6, and TNF-α. It has been investigated in various clinical applications, including transdermal pain management (e.g., with oxycodone), topical treatments for acne, and oral delivery of nutrients and pharmaceuticals.
