Tocopheryl phosphate mixture (TPM) is a proprietary, lipid-based drug delivery technology developed by Avecho Biotechnology (formerly Phosphagenics). It is composed of a mixture of two phosphorylated forms of vitamin E: mono-tocopheryl phosphate (TP) and di-tocopheryl phosphate (T2P). TPM self-assembles into vesicular nanostructures that encapsulate active pharmaceutical ingredients (APIs), enhancing their solubility, stability, and absorption across dermal, transdermal, oral, and injectable routes. In addition to its role as a penetration enhancer and delivery vehicle, TPM possesses inherent anti-inflammatory and skin-soothing properties. It has been utilized in various clinical-stage formulations, including a transdermal patch for oxycodone (evaluated in Phase II trials for postherpetic neuralgia and pain) and an oral softgel capsule for cannabidiol (CBD) (in Phase III trials for insomnia).
