TOL-2506 is an extended-release formulation of leuprolide acetate, a gonadotropin-releasing hormone (GnRH) agonist, developed by Tolmar for oncology indications. It is specifically being investigated for ovarian function suppression in premenopausal women and men with hormone receptor-positive (HR+), HER2-negative breast cancer. Formulated as a long-acting injectable suspension for subcutaneous administration every 12 weeks, TOL-2506 works by continuously stimulating GnRH receptors in the pituitary gland, which leads to their downregulation and a subsequent decrease in the secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). This suppression results in reduced levels of estrogen and testosterone, which are drivers in certain types of breast cancer. The drug is currently being evaluated in the Phase 3 OVELIA clinical trial.
