**TolAPC** is a preclinical tolerogenic artificial antigen-presenting-cell microparticle developed at Johns Hopkins University for antigen-specific treatment of autoimmune disease. The reported multiple-sclerosis construct comprises a poly(lactic-co-glycolic acid) and poly(beta-amino ester) microparticle loaded with rapamycin and surface-conjugated with an interleukin-2 immunocytokine and MHC class II tetramers presenting a myelin oligodendrocyte glycoprotein peptide. It is designed to preferentially expand regulatory T cells and tolerize myelin-reactive T cells. In experimental autoimmune encephalomyelitis, a mouse model of multiple sclerosis, prophylactic dosing delayed disease onset and reduced severity, while therapeutic dosing reversed symptomatic disease and increased regulatory T cells in the central nervous system.
