Overview

Tolciclate is a small molecule antifungal agent belonging to the thiocarbamate class. It is primarily used topically for the treatment of dermatophyte infections caused by fungi such as Epidermophyton, Microsporum, and Trichophyton species. Its mechanism of action involves inhibition of fungal sterol biosynthesis by blocking squalene epoxidase in the ergosterol pathway. This leads to accumulation of squalene and disruption of fungal cell membrane synthesis. Tolciclate exhibits similar in vitro activity to tolnaftate but has greater liposolubility and higher efficacy in animal models[1][3][5][6][8].
