Tolero-LNPs is a lipid nanoparticle (LNP)-based RNA medicine delivery platform developed by ISAR Bioscience, a Bavarian research company. The platform utilizes innovative, patent-protected lipids that are designed to be biodegradable and exceptionally well-tolerated by the immune system. It is specifically engineered to deliver mRNA payloads that generate immune tolerance, making it a potential therapeutic modality for autoimmune disorders. The lead application is the treatment of Graves' disease (affecting both the thyroid and the eyes), where the platform has demonstrated the ability to modulate dendritic cell activation in patient-derived samples. As of late 2024, the technology is in the preclinical development stage, with ongoing GMP manufacturing and toxicological studies aimed at initiating clinical trials.
