Overview

Tolerogenic dendritic cell therapy (RASGRP2 peptide-based) is an autologous cell therapy under development for the treatment of multiple sclerosis (MS). The therapy utilizes patient-derived dendritic cells that are treated with tolerogenic stimuli and loaded with immunogenic peptides derived from RAS guanyl-releasing protein 2 (RASGRP2), a calcium- and diacylglycerol-regulated nucleotide exchange factor identified as a major autoantigen in HLA-DRB1*15:01 (DR15)-positive MS patients. By presenting these cognate autoantigens in a tolerogenic context, the therapy aims to re-establish antigen-specific immune tolerance, selectively suppressing the pathogenic, autoreactive CD4+ T-cell responses that drive demyelination in MS without causing broad systemic immunosuppression.
