**Tolerogenic microspheres** are a preclinical microsphere-based immunotherapy for **type 1 diabetes** developed from work at the University of Pittsburgh and associated with DiaVacs. The formulation encapsulates antisense oligodeoxynucleotides directed against **CD40, CD80, and CD86**, reducing dendritic-cell costimulatory capacity and promoting a tolerogenic dendritic-cell phenotype. In non-obese diabetic mouse studies, administration generated antigen-specific Foxp3-positive regulatory T cells in draining pancreatic lymph nodes, reduced autoreactive immune responses to beta-cell antigens, and prevented or reversed hyperglycemia without broad systemic immunosuppression.
