Overview

Tolisign is a brand name owned by Primus Remedies for a line of combination oral therapies containing the centrally acting muscle relaxant tolperisone. The product line includes Tolisign-P, a fixed-dose combination of tolperisone (150 mg) and paracetamol (325 mg), and Tolisign-D, a fixed-dose combination of tolperisone (150 mg) and diclofenac (50 mg). Tolperisone acts as a muscle relaxant by blocking voltage-gated sodium and calcium channels in the spinal cord and brainstem, thereby inhibiting mono- and polysynaptic spinal reflexes. Paracetamol and diclofenac provide analgesic and anti-inflammatory effects by inhibiting cyclooxygenase (COX) enzymes (prostaglandin G/H synthases). These combinations are indicated for the treatment of painful muscle spasms and acute musculoskeletal conditions.
