TOLISIGN-P is a fixed-dose combination oral medication developed and marketed by Primus Remedies, containing the centrally acting muscle relaxant tolperisone and the analgesic paracetamol (acetaminophen). Tolperisone exerts its muscle relaxant effect by inhibiting voltage-gated sodium and calcium channels in the spinal cord and brainstem, which stabilizes nerve membranes and suppresses spinal reflex activity without causing significant sedation. Paracetamol provides analgesic and antipyretic effects primarily through the inhibition of cyclooxygenase (COX) enzymes in the central nervous system, thereby reducing prostaglandin synthesis. TOLISIGN-P is indicated for the treatment of pain associated with muscle spasms, providing a dual-action approach to alleviate both the muscle hypertonicity and the accompanying pain.
