Tolododekin alfa (ANK-101) is a first-in-class anchored drug conjugate in which interleukin-12 (IL-12), a cytokine with potent antitumor activity, is stably linked to aluminum hydroxide via an alum-binding peptide. This design allows for localized and prolonged IL-12 activity within the tumor microenvironment while minimizing systemic toxicity. Tolododekin alfa is being developed as an intratumoral immunotherapy for advanced solid tumors, aiming to enhance immune-mediated tumor destruction and improve responses to chemotherapy and immune checkpoint inhibitors. Preclinical studies have demonstrated durable antitumor effects after a single dose in animal models, with favorable safety profiles observed across multiple species. The drug is currently undergoing Phase 1 clinical trials in patients with advanced solid tumors[1][2][3][7].
