Tolododekin alfa (ANK-101) is an anchored interleukin-12 (IL-12) immunotherapy designed for intratumoral administration. It consists of a recombinant human IL-12 (tolododekin alfa) conjugated to an anchoring moiety that binds to the extracellular matrix (ECM) within the tumor microenvironment. This anchoring mechanism is intended to prevent systemic leakage of IL-12, thereby reducing systemic toxicity while maintaining high local concentrations to stimulate an anti-tumor immune response. In combination with anti-PD-1/PD-L1 antibodies, such as the PD-1 inhibitor cetrelimab, the therapy aims to overcome the immunosuppressive tumor microenvironment and enhance T-cell mediated anti-tumor activity. The combination is currently being evaluated in Phase 1 clinical trials for patients with advanced or metastatic solid tumors, including non-small cell lung cancer (NSCLC) that has progressed after initial treatments.
