Overview

Tolonidine is an antihypertensive drug that belongs to the class of centrally acting alpha-2 adrenergic agonists, structurally related to clonidine. It acts by stimulating presynaptic alpha-2 adrenergic receptors in the central nervous system, leading to decreased sympathetic outflow and reduced blood pressure. Tolonidine was developed as an alternative to clonidine for the treatment of hypertension but is not widely used or marketed in most countries. Its mechanism of action involves reducing norepinephrine release from sympathetic nerve terminals, resulting in vasodilation and lowered heart rate.
