Tomligisiran is an investigational, liver-targeted **GalNAc-conjugated small interfering RNA** developed as one constituent of the fixed siRNA combination daplusiran + tomligisiran, formerly JNJ-3989 and ARO-HBV. It is designed to trigger RNA interference against hepatitis B virus X open reading frame messenger RNA in cccDNA-derived HBV transcripts, reducing viral RNA translation and thereby suppressing HBV protein production and replication. The daplusiran + tomligisiran combination is being evaluated by GSK in sequential treatment regimens with bepirovirsen for chronic hepatitis B.
