Overview

Tomuzotuximab (formerly known as CetuGEX) is a glyco-optimized, humanized monoclonal antibody targeting the epidermal growth factor receptor (EGFR). Developed by Glycotope using its proprietary GlycoExpress™ platform, the antibody is engineered to have an optimized glycosylation pattern, specifically lacking fucose, which significantly enhances its affinity for the FcγRIIIa receptor on immune effector cells. This modification is intended to increase antibody-dependent cellular cytotoxicity (ADCC) compared to first-generation EGFR inhibitors like cetuximab. Despite its enhanced immune-recruiting potential, a Phase II trial (RESGEX) in patients with recurrent or metastatic squamous cell carcinoma of the head and neck (SCCHN) showed that tomuzotuximab combined with chemotherapy did not provide a progression-free survival benefit over cetuximab.
