TOP-V122

TOP-V122 is a dual-mode small molecule drug candidate developed by Topadur Pharma AG. It acts as both a potent and selective phosphodiesterase 5A (PDE5A) inhibitor and a nitric oxide (NO) donor. This combination restores intracellular cGMP levels, leading to improved microcirculation, healthy angiogenesis, neuroprotection, and inhibition of hypoxia. The drug is being developed primarily for ophthalmic indications such as dry age-related macular degeneration (AMD), non-proliferative diabetic retinopathy (DR), and normal pressure glaucoma. Additional preclinical studies are ongoing for colorectal cancer and pulmonary fibrosis. Formulations under development include eye drops for monotherapy in early-stage disease and injectable sustained-release forms for use alongside anti-VEGF drugs in more severe cases[1][2][6].
