Topfond Pharma-DCN1-UBC12-small molecule is an undisclosed preclinical-stage small molecule developed by Topfond Pharma (Henan Topfond Pharmaceutical). The compound is designed to target the protein-protein interaction between Defective in Cullin Neddylation 1 (DCN1) and the ubiquitin-conjugating enzyme E2 M (UBC12, also known as UBE2M). This interaction is a critical step in the neddylation pathway, which activates Cullin-RING ligases (CRLs). CRLs are the largest family of E3 ubiquitin ligases and are frequently overexpressed in various cancers, where they promote the degradation of key tumor suppressors. By inhibiting the DCN1-UBC12 interface, this small molecule prevents Cullin neddylation, leading to the stabilization of tumor suppressor proteins and the inhibition of cancer cell growth. It is currently being investigated as a potential targeted therapy for oncology indications.
