Overview

Topical aminocaproic acid (SEI003) is a small molecule serine protease inhibitor being developed by the University of California, San Diego, for the treatment of papulopustular rosacea. While aminocaproic acid is FDA-approved for systemic use in hemostasis, this specific 25% topical formulation aims to inhibit Kallikrein 5 (KLK5). KLK5 is a trypsin-like serine protease that, when overactive, leads to the production of the pro-inflammatory antimicrobial peptide LL-37 (cathelicidin), a key driver of rosacea symptoms. By reducing KLK5 activity and subsequent LL-37 levels, the drug aims to alleviate skin inflammation.
