Topical lingual amylin is an investigational formulation of the hormone amylin, designed for delivery via the tongue (lingual route) as a topical agent. Amylin is a 37-amino acid peptide hormone co-secreted with insulin by pancreatic beta cells in response to food intake. It acts on satiety centers in the brain to reduce food intake, delay gastric emptying, and inhibit glucagon secretion, thereby improving glucose metabolism and promoting weight loss. The topical lingual approach leverages neural signaling between the tongue and brain to activate satiety pathways without systemic exposure, potentially reducing side effects associated with injectable or systemic therapies. This modality is being developed primarily for obesity and cardiometabolic disorders[1][7].
