topical romidepsin

Topical romidepsin is an ointment formulation of the histone deacetylase (HDAC) inhibitor romidepsin, being developed by the National Cancer Institute (NCI) for the treatment of early-stage cutaneous T-cell lymphoma (CTCL). While the intravenous formulation (Istodax) is FDA-approved for relapsed/refractory CTCL, this skin-directed therapy aims to treat patients with stage IA, IB, or IIA disease. By inhibiting HDAC enzymes, romidepsin regulates gene expression to induce cell cycle arrest and apoptosis in malignant T cells. The topical application is intended to provide localized efficacy while potentially minimizing the systemic toxicity associated with intravenous administration.
