Topical sodium nitrite is an investigational 2% cream formulation developed by Dr. Caterina P. Minniti and colleagues at the National Heart, Lung, and Blood Institute (NHLBI) for the treatment of chronic leg ulcers, particularly in patients with sickle cell disease. As a nitric oxide donor, sodium nitrite promotes vasodilation, thereby increasing peri-wound blood flow and oxygenation to the ulcer bed. Additionally, it exhibits bacteriostatic properties that may help reduce the bacterial load in chronic wounds. Clinical studies, including a Phase 2 trial funded by the FDA Office of Orphan Products Development, have evaluated its safety and efficacy in accelerating ulcer healing and reducing associated pain, with promising results observed in ulcers of shorter duration.
