Topical STAT6 is a preclinical-stage small molecule inhibitor of Signal Transducer and Activator of Transcription 6 (STAT6), developed by LEO Pharma in collaboration with Gilead Sciences. STAT6 is a critical transcription factor in the IL-4 and IL-13 signaling pathways, which are central drivers of Type 2 inflammation. This pathway is highly implicated in the pathophysiology of various inflammatory skin conditions, most notably atopic dermatitis. By delivering the inhibitor topically, the treatment aims to provide localized anti-inflammatory effects within the skin while minimizing the potential for systemic side effects often associated with oral or injectable JAK/STAT inhibitors.
