Overview
Topirazumab (also known as VDJ-001) is a humanized monoclonal antibody that acts as an antagonist of the insulin-like growth factor-1 receptor (IGF-1R). Developed by Beijing Vantage Biopharma, it is primarily being investigated for the treatment of thyroid-associated orbitopathy (TAO), also known as thyroid eye disease. Similar to the mechanism of teprotumumab, topirazumab binds to IGF-1R and blocks its activation, which is upregulated in the orbital fibroblasts of patients with TAO. By inhibiting IGF-1R signaling, the drug aims to reduce the inflammatory response, glycosaminoglycan production, and adipogenesis that drive orbital tissue expansion and clinical symptoms such as proptosis (eye bulging) and diplopia. As of 2024, topirazumab is in Phase II clinical development in China.
