Topofen is a proprietary topical gel formulation containing 5% ketoprofen, a potent nonsteroidal anti-inflammatory drug (NSAID). Developed by Achelios Therapeutics using their Achetogel delivery platform, Topofen is designed for rapid and efficient skin permeation to treat pain locally while minimizing systemic exposure. Its primary mechanism involves inhibition of prostaglandin synthesis via cyclooxygenase and lipoxygenase pathways, leading to reduced neurogenic inflammation. Topofen has demonstrated efficacy in clinical studies for the acute treatment and prevention of migraine (including menstrual and episodic forms) as well as chronic temporomandibular joint pain. The drug’s effect is mediated through regulation of neuronal CGRP release by suppressing prostaglandin generation at trigeminal nerve endings[1][2][3][7].
