Overview

Toripalimab is a humanized IgG4 monoclonal antibody that functions as an immune checkpoint inhibitor targeting the programmed cell death protein 1 (PD-1) receptor. By binding to PD-1, it blocks its interaction with ligands PD-L1 and PD-L2, thereby preventing the inhibition of T-cell activation and restoring anti-tumor immune responses. Toripalimab has demonstrated high affinity for PD-1 and promotes robust Th1-type immune responses. It is primarily indicated for the treatment of nasopharyngeal carcinoma (NPC), both as monotherapy in patients with recurrent or metastatic disease after platinum-based chemotherapy, and in combination with gemcitabine and cisplatin as first-line therapy for metastatic or recurrent locally advanced NPC. The drug was first approved in China for melanoma and received FDA approval in October 2023 for NPC.
