Toripalimab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, developed by Shanghai Junshi Biosciences. In a clinical trial context led by Ruijin Hospital, it is being investigated as an induction therapy in combination with a chemotherapy regimen consisting of 5-fluorouracil and cisplatin (or carboplatin) for patients with locoregionally-advanced laryngo-hypopharyngeal squamous cell carcinoma. The mechanism of action involves toripalimab blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2) to restore anti-tumor T-cell activity, while the chemotherapy components provide cytotoxic effects by inhibiting DNA synthesis and inducing DNA damage. This combination approach is intended to reduce tumor burden and improve organ preservation rates in patients with locally advanced laryngeal and hypopharyngeal cancers.
