Overview

toripalimab + actinomycin-d is a combination therapy consisting of toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and actinomycin-D (also known as dactinomycin), a cytotoxic chemotherapy agent. This regimen is being evaluated as a first-line treatment for patients with high-risk gestational trophoblastic neoplasia (GTN), specifically those with a FIGO score of 7. Toripalimab acts as an immune checkpoint inhibitor to enhance the body's anti-tumor immune response, while actinomycin-D inhibits transcription by intercalating into DNA and interfering with RNA polymerase. The combination is currently under investigation in a Phase II clinical trial led by Peking Union Medical College Hospital to assess its efficacy in achieving complete response and its safety profile in this patient population.
