Overview

Toripalimab + anlotinib + chemotherapy is a combination regimen used in oncology, particularly for advanced or metastatic cancers. Toripalimab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, functioning as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor responses. Anlotinib is a small molecule multi-targeted tyrosine kinase inhibitor that primarily inhibits vascular endothelial growth factor receptors (VEGFR), fibroblast growth factor receptors (FGFR), platelet-derived growth factor receptors (PDGFR), and c-Kit, thereby exerting anti-angiogenic and anti-proliferative effects. Chemotherapy refers to cytotoxic agents commonly used in cancer treatment; specific agents may vary by protocol but typically include drugs such as platinum-based compounds or anthracyclines. This combination has shown promising efficacy and manageable safety profiles in several cancer types including esophageal squamous cell carcinoma, nasopharyngeal carcinoma, soft tissue sarcoma, biliary tract cancer, hepatocellular carcinoma, and small-cell lung cancer. The rationale for combining these agents lies in their complementary mechanisms—anlotinib can normalize tumor vasculature and modulate the tumor microenvironment to enhance immune infiltration and response to toripalimab; chemotherapy can further reduce tumor burden and potentially increase immunogenicity[1][2][5].
