This combination therapy consists of the PD-1 inhibitor toripalimab and the chemotherapy agents docetaxel and cisplatin. It is being investigated as induction chemotherapy for patients with localized or locally advanced hypopharyngeal squamous cell carcinoma (HPSCC). Toripalimab is a monoclonal antibody that blocks the interaction between the PD-1 receptor and its ligands (PD-L1 and PD-L2), thereby enhancing the immune system's ability to recognize and attack tumor cells. Docetaxel is a taxane that stabilizes microtubules, preventing their depolymerization and leading to cell cycle arrest. Cisplatin is a platinum-based alkylating agent that forms DNA cross-links, interfering with DNA repair and synthesis. The regimen is being evaluated by the Peking Union Medical College Hospital to improve laryngeal preservation rates and reduce tumor burden prior to surgery or radical chemoradiotherapy.
