This combination therapy consists of toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, administered alongside the chemotherapy agents docetaxel and cisplatin. Toripalimab functions as a checkpoint inhibitor, blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2) to restore T-cell mediated anti-tumor activity. Docetaxel is a taxane that promotes microtubule assembly and inhibits disassembly, leading to cell cycle arrest, while cisplatin is a platinum-based agent that forms DNA cross-links, interfering with DNA replication and transcription. This triplet regimen is being investigated as induction chemotherapy for localized hypopharyngeal squamous cell carcinoma (HSCC) to improve organ preservation rates and reduce tumor burden prior to definitive surgery or chemoradiotherapy.
