Overview

Toripalimab + gemcitabine is a combination therapy consisting of toripalimab, a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), and gemcitabine, a nucleoside analog used as chemotherapy. Toripalimab blocks the PD-1 receptor on T cells to enhance immune-mediated tumor cell killing by preventing interaction with its ligands PD-L1 and PD-L2. Gemcitabine inhibits DNA synthesis by incorporating into DNA during replication, leading to apoptosis in rapidly dividing cancer cells. This combination has been studied primarily for the treatment of recurrent or metastatic nasopharyngeal carcinoma (NPC), especially in patients who are ineligible for cisplatin-based regimens or as part of first-line therapy. Clinical trials have shown that this regimen offers favorable response rates and improved survival outcomes with manageable toxicity profiles[1][2][3].
